Determination of thyroxine-binding globulin in human serum by single radial immunodiffusion and radioimmunoassay.

چکیده
Two immunochemical methods for determination of thyroxine-binding globulin in human serum were developed, in which the purified globulin and monospecific antiserum to it are used. One method, based on radial immunodiffusion, has good precision and values for analytical recovery. Reference values obtained for men were 9.8-17.8 mg/liter and for women 11.3-20.5 mg/liter. The sex-related difference was significant. The other method is based on radioimmunoassay, with use of an iodinated acylating agent for the labeling of thyroxine-binding globulin. The relative merits of the two methods are discussed.

Serum free thyroxine and free triiodothyronine concentrations in pregnancy.
Blood was taken from women in each trimester of pregnancy at routine antenatal visits. Further samples were collected from an age matched control group of euthyroid women taking no medication. Free triiodothyronine and free thyroxine concentrations were measured by direct radioimmunoassay (Amerlex Kit methods, Amersham International, Amersham, Buckinghamshire).2 These free hormone assays use th...
متن کاملSerum Alpha-1-antitrypsin Evaluation by Three Electrophoretic, Enzymatic and Immunodiffussion Methods
Abstract Background & Objective: Alpha-1-antitrypsin (AAT) is the major component of the human plasma alpha-1 globulin proteins and acts as a major inhibitor of proteolytic enzymes, particularly elastase. AAT deficiency is accompanied by lung, liver and other disorders, therefore, AAT is clinically important and its precise evaluation is diagnostically critical. In present study serum AAT was e...
متن کاملRadioimmunoassay for human thyroxine-binding prealbumin.
A radioimmunoassay (RIA) for human thyroxine-binding prealbumin (PA) is described. It employs highly purified PA, anti-human PA serum at 1:30,000 final dilution, normal bovine serum as a carrier, and polyethyleneglycol to precipitate the immune complexes. This assay is extremely sensitive (limit of detection less than 0.2 micrograms per dL or less than 3.6 X 10(-15) moles per tube), accurate (r...
متن کاملRadioimmunoassay for serum triiodothyronine: evaluation of simple techniques to control interference from binding proteins.
Simple techniques for controlling interference from binding proteins in serum, such as thyroxine-binding globulin, in radioimmunoassay for triiodothyronine (13) have been evaluated for their efficacy, and their effect on assay sensitivity and on recovery of added T3. Ethanol precipitation of serum proteins decreased the assay sensitivity, nonspecific binding was increased, and recoveries of add...
متن کاملInterference of iodine-125 ligands in radioimmunoassay: evidence implicating thyroxine-binding globulin.
Although there is abundant published evidence that radioiodinated antigens interfere in digoxin radioimmunoassays, other radioimmunoassays are similarly affected. We investigated the relationship of radioiodinated antigen structure to its function in the immunoassay. Carrier-free 125I-labeled iodotyrosine and iodohistamine derivatives were incubated with human serum, and the bound and free frac...
